Schwerpunkte:

Die Arbeitsgruppe beschäftigt sich vorrangig mit den Ursachen des Untergangs von Synapsen bei Neurodegenerativen Erkrankungen wie dem Morbus Alzheimer, den Prionerkrankungen und dem Morbus Parkinson. Dazu wird zum einen anhand von Knock-out Mäusen die normale Funktion der Proteine untersucht, die bei diesen Erkrankungen Schlüsselpositionen einnehmen, wie das Amyloid Vorläufer Protein (APP), die Preseniline, BACE, Alpha-Synuclein oder das Prionprotein (PrP). Zum anderen werden im Detail die einzelnen Schritte des synaptischen Schadens anhand unterschiedlicher transgener Mausmodelle dieser Erkrankungen untersucht.

Neben klassischen molekulargenetischen, biochemischen, elektrophysiologischen und histologischen Methoden haben wir uns auf die in vivo Zweiphotonenmikroskopie spezialisiert. Diese Methode erlaubt es, über Wochen bis Monate den Krankheitsverlauf in vivo direkt zu beobachten und z.B. den Effekt von neuen therapeutischen Ansätzen, die den Untergang von Synapsen aufhalten sollen, direkt zu verfolgen. Im Rahmen von primär technischen Projekten arbeiten wir des Weiteren an der Weiterentwicklung der Zweiphotonenmikroskopie und sehr erfolgreich an dem Einsatz dieser Methode im Rahmen von neuro-onkologischen Projekten in Zusammenarbeit mit der AG-Winkler.
